Scope of aims: This is an introductory course to introduce the knowledge and skills of various cryo TEM/SEM sample preparation techniques, including high pressure freezing, plunge freezing and cryo-ultramicrotomy
Targeted participants: Individuals who wish to learn cryo EM sample preparation of biological specimen. No experience is required.
Format: Lecture, demonstration, and hands-on practice
Techniques covered: Cryo-immobilization, High Pressure freezing, Automated freeze substitution, Cryo-ultramicrotomy, Grid plunging, Rapid freeze substitution, etc.
Instrumentation: High pressure freezer ICE and EMPACT II (Leica-Microsystems), AFS (Leica-Microsystems), Grind plunger (Leica-Microsystems), Cryo-ultramicrotome UC6/FC7 (Leica-Microsystems)
